I complete the run on emulator (Puyo Puyo Tsu REV00) in roughly 8 minutes 36 seconds, from the moment I select the single player mode until the fade out after I beat Masked Satan (Unlockable hidden boss if you reach the last stage with over 150k points and have never lost a continue). The in-game time is 406 seconds, or 6 minutes and 46 seconds of gameplay.

In order to achieve 400k points, I manipulate the RNG at the start by setting it to a known value at the beginning of stage 1. Its value changes every small timestep during the intro and in the main menu, so by mashing the start and a,b,c buttons until the tower shows up, I make sure that I get a specified distribution of colors against the first opponent (for the later opponents other parameters interfere and make it hard to predict). Since it depends on which character I am pitted against as well, I continue mashing so I get Skeleton-T. Then I know exactly what to do in order to get that 16 chain in stage 1, landing me 295k points. Then I just have to survive the later stages of the game while scoring decently in order to get the remaining 105k points. Things go smoothly here but the RNG can make a hell out of this game !
